The PD-L1 Stain Kit is intended for laboratory use in the qualitative detection of programmed death ligand 1 (PD-L1) protein via immunofluorescence (IF). This protocol utilizes a biotin-conjugated PDL1 aptamer followed by Alexa Fluor™ 488-conjugated Streptavidin for signal visualization.
The PD-L1 aptamer specifically recognizes the PD-L1 protein and is suitable for immunofluorescence staining of cells to assess PD-L1 expression in the cell membrane or cytoplasm.
Key Features and Advantages
Compared to antibodies, this aptamer-based kit offers the following advantages:
• Versatile Staining Options: The kit is compatible with both fixed and live cells, providing
flexibility for various experimental designs.
• Dynamic Live Staining Mechanism: When used for live cell staining, the aptamer can be
processed via cellular metabolism and transported to the cell surface, enabling unique observation
of cellular dynamics.
• High Specificity and Sensitivity: Ensures accurate detection with high affinity.
• Consistency: Minimal batch-to-batch variability and excellent reproducibility.

Preparation And Storage
1. PD-L1 Aptamer
Please refer to the vial label and reconstitute in nuclease-free water at the concentration of 100 µM. For long-term storage (1 year), aliquot and store at -20ºC. Avoid multiple freeze-thaw cycles. Recommended dilution 1:100 - 1:500.
Pre-staining process: Apply PD-L1 Aptamer (100 µM) in Aptamer Dilution Buffer at 1:100 to 1:500. Heat at 95°C for 10 minutes, then immediately place on ice for 10 minutes.
